Assessment of circulating 25(OH)D and 1,25(OH)2D: emergence as clinically important diagnostic tools.

چکیده
One of the major factors responsible for the explosion of knowledge related to vitamin D metabolism and its relation to clinical disease was the introduction of competitive protein-binding assays for 25(OH)D and 1,25(OH)2D. 2 These assays were introduced more than three decades ago, and were based on assessing circulating 25(OH)D or 1,25(OH)2D using the vitamin D-binding protein or chick intestinal receptor, respectively, as the primary binding agents. Both of these assays used H-labeled compounds as reporters. Although these assays were valid, they were also relatively cumbersome, especially the 1,25(OH)2D procedure. Through the years, assays for both of these metabolites have advanced. Some advances have been for the better, some have not. These methodologies include radioimmunoassay (RIA), enzyme-linked immunoassay (ELISA), high-performance liquid chromatography (HPLC), liquid chromatography coupled with mass spectrometry (LC-MS), and random access automated assay (RAAA) based on chemiluminescence assay technology.

Vitamin D deficiency: consequence or cause of CKD?
Pioneering studies in the 1970s demonstrated that CKD leads to substantial perturbations in the vitamin D endocrine system and, as a result, bone loss (1–4). In particular, in CKD, renal production of 1,25-dihydroxyvitamin D (1,25(OH)2D) from 25hydroxyvitamin D (25(OH)D) is markedly reduced, leading to low circulating 1,25(OH)2D concentrations. Insufficient 1,25(OH)2D contributes to the develop...
متن کاملThe interrelationship of serum 1,25-dihydroxyvitamin D, 25-hydroxyvitamin D and 24,25-dihydroxyvitamin D in pregnancy at term: a meta-analysis.
OBJECTIVE To assess the relationship between circulating 25(OH)D, 1,25(OH)2D, and 24,25(OH)2D in pregnancy at term, and to test the hypothesis that serum 25(OH)D and 24,25(OH)2D are repressed by high serum 1,25(OH)2D levels. DESIGN Twenty studies in pregnant women at term and 14 studies in young non-pregnant women were selected from the electronic literature. From these studies the mean serum...
متن کاملAssessment and interpretation of circulating 25-hydroxyvitamin D and 1,25-dihydroxyvitamin D in the clinical environment.
The unique cis-triene structure of vitamin D and related metabolites makes it susceptible to oxidation, ultraviolet (UV) light-induced conformational changes, heat-induced conformational changes, and attacks by free radicals. Vitamin D(2) is much less bioactive than vitamin D(3) in humans. Metabolic activation and inactivation of vitamin D are well characterized and result in a plethora of meta...
متن کاملThe TaqI gene polymorphisms of VDR and the circulating 1,25-dihydroxyvitamin D levels confer the risk for the keloid scarring in Chinese cohorts.
AIM to investigate the association between vitamin D receptor polymorphisms and circulating 1,25-Dihydroxyvitamin D (1,25(OH)2D) levels with keloid scar (KS) risk. METHODS A total of 261 patients with KS and 261 normal healthy individuals were enrolled. VDR gene polymorphisms were determined. Circulating 1,25(OH)2D levels were detected. RESULTS In this study, we investigated the role of fou...
متن کاملA Prospective Study of Plasma Vitamin D Metabolites, Vitamin D Receptor Polymorphisms, and Prostate Cancer
BACKGROUND Vitamin D insufficiency is a common public health problem nationwide. Circulating 25-hydroxyvitamin D3 (25[OH]D), the most commonly used index of vitamin D status, is converted to the active hormone 1,25 dihydroxyvitamin D3 (1,25[OH]2D), which, operating through the vitamin D receptor (VDR), inhibits in vitro cell proliferation, induces differentiation and apoptosis, and may protect ...
